This quest occurs through the Companions quest line.

Talk to Vilkas and choose to continue the quest to make him a lover, or not (if guys aren't your bag). The quest will end immediately and you'll miss all his custom dialogue if guys are not your thing.

Talk to him again after completing Quest C01 - Proving Honor

Talk to him again after completing Quest C03 - The Silver Hand

Talk to him again after completing Quest C06 - End of the companions quest line.

Quest completes and he becomes a lover.

STAGE 10 - I have met Vilkas of the Companions. A strong and noble warrior. He seems a bit reserved. Life in Skyrim usually does not wait for long courtships, however.

STAGE 20 - I will need to choose if trying to get Vilkas as a lover is right for me.

STAGE 25 - I will need to join the Companions and earn the respect and trust of Vilkas if I want to find a way to his heart. I should perform missions and seek further dialogue opportunities.

STAGE 30 - Vilkas seems like he is not interested in a relationship. I should continue working for the Companions and try talking to him again from time to time. Maybe he will open up.

STAGE 40 - The nerve of Vilkas, assuming I would continue to pursue him after he's rebuffed my advances. He's got another thought coming.

STAGE 50 - Vilkas was concerned about the Beast Blood within himself after a girl he liked was killed by one. It has made him distant. In the end he sought out my affections and now we are lovers.

STAGE 199 - Yeah... I'm just not into having men as lovers.

STAGE 200 - Shut down Quest, Mark Completed
