Must I run FNIS after creating a new MO profile?

When in doubt, run it. That's what I always do anyway.

 

You're absolutely right on that notion, although it would be nice to know if running FNIS again is necessary.

 

 

Since I don't use MO, I don't know how it works, but if it involves disabling/enabling mods that include new animations, you'd want to rerun FNIS

After creating a new MO profile, is it necessary to run FNIS again even if all your mods are automatically disabled?

 

Assuming you mean on the left hand side, yes you will have to run FNIS again. If you are talking about the right hand side, no. Basically run FNIS if you enable/disable any mod on the left hand side which has animations or a skeleton or generally anything what FNIS deals with.

Left hand side mod packages, (including esp's, esm's,  textures, scripts, etc.). Right hand side the .esp's & .esm's which would normally be in your data directory, (obviously these may or not be enabled). Simplified because what is enabled in the left hand side is reflected in the virtual data directory the right hand side represents, (i.e. including textures & scripts), but in the order that you have it in the left hand side for textures and scripts, but probably if using LOOT not for esp's and esm's. (Actually probably making this sound more complicated than it is). So yes to your clarification.
